Two Rapid Assays for Saponin in Individual Alfalfa Plants1

Two assays were developed for screening individual alfalfa (Medicago saliva) plants for crude saponin. Twenty alfalfa leaf samples, selected on the basis of erythrocyte assay results, were evaluated by the fish assay and secondly by a chemical determination for saponins. The correlation coefficients of these assay results were ∝erythrocyte vs. chemical (r = .90), √fish vs. √erythrocyte (r = −.91), and √fish vs. chemical (r = −.81). Results indicate that the two assays may provide rapid and accurate estimates of crude saponin in alfalfa plants.
